In a small clinical trial conducted in the United States, researchers tested an experimental oral vaccine called WRSs2, which aims to protect against shigellosis, a disease caused by the Shigella bacterium. The trial involved 108 adults aged 18 to 49 who had little preexisting immunity to the Shigella species S. sonnei. Participants received two doses of the vaccine, which contained a weakened strain of the bacteria. The results indicated that the vaccine was effective, as only 3 out of 34 vaccinated participants developed shigellosis after being exposed to a live, full-strength dose of the bacteria, compared to 21 out of 26 participants who received a placebo. Furthermore, only 3 percent of vaccinated individuals experienced severe symptoms, while 69 percent of placebo recipients did. Despite these encouraging results, experts caution that the vaccine is still far from approval, as larger clinical trials are necessary to confirm its effectiveness and safety. The research highlights the urgent need for effective vaccines against shigellosis, especially given that the bacterium causes an estimated 450,000 infections annually in the U.S. and 80 million infections worldwide. The study's findings also raise questions about the vaccine's applicability to different populations, particularly young children in low- and middle-income countries, who are most affected by the disease. Researchers plan to continue testing WRSs2 and aim to evaluate its effectiveness in populations that are most vulnerable to shigellosis.
